The Greens have started the season with wins over the Black and Whites and Yenda, while the Mallee Men knocked off the Blueheelers in round one before having the bye last weekend.
Leeton coach Hayden Philp feels it will be a great test to see where his side is sitting in the early stages of the season.
"They play very similar to us, very quick footy, and they will definitely be one of our tougher contests throughout the year," he said.
"We played Black and Whites first round, and they are only going to get better and better each week but to play a side like Wyalong who is always a very good side and play very high skilled footy.
"It will be good to see where we are actually at."
Philp knows his side will need to hold onto the ball much better than they did against Yenda, having won the game despite having a poor competition rate.
"We were probably lucky to win games like that because I think it was less than 50 per cent for the game," he said.
"The score probably didn't reflect how tight the contest actually was, but if we can complete at least 60 or 70 per cent, those points probably would have been a bit more. It was full credit to the boys for being able to defend the way they did.
"They only scored four points with 12 blokes on the field in the last five minutes, so it was a massive effort."

This weekend will see the Greens without the services of George Broome, and there is a bit of a wait and see on a couple of others.
Meanwhile, West Wyalong will be without their captain-coach, Jake Goodwin, who will represent Country against City this weekend.
For Goodwin, the selection came as a surprise, but it will be a chance to follow in his father's footsteps.
"It is really humbling to be picked in that side and to be wearing the same jersey as my old man did when he was playing for West Wyalong," he said.
"It did a bit because I wasn't really watching the other teams, so I was a bit surprised. I didn't really know what the calibre was like with the other teams. I just wanted to play good footy for Riverina, and then this came about."
It will cause a bit of a reshuffle for the Mallee Men, but Goodwin fees his club has the depth to deal with the challenge.
"Our depth is pretty good, so I'm looking forward to seeing how the boys go in different positions," he said.
" At some stages, it has been a bit shaky. We don't have 16s, 18s are pretty tight with numbers, but for first and reserve grade, we have been pretty good.
"The more people that are turning up, the more are getting on board. After having the win in round one, a lot of people are getting behind us a bit more."
The West Wyalong side played five debutants in their win over Yenda, and Goodwin said it was great to see those boys make the step up.
The clash between the ladder leaders will get underway at 3pm at Leeton No 1 Oval.
