They'll be taking in the scenic view from the waters of the Murrumbidgee as west as part of the Redneck River Run.
The run is a fundraising event for Country Hope, and is a sister event to the Riverina Redneck Rally.
The boats will cover 40 kilometres each day and event organiser Gavin Gilbert expected the fleet would be reaching the Carrathool bridge on Sunday before arriving in Hay on Wednesday.
So far 90 people for 40 boats have entered.
As well as supporting Country Hope, Mr Gilbert said the sailors would be showcasing the communities along the river to people from Coffs Harbour, Newcastle and beyond.
"The amount of interest we've had means we're going to make it an annual event," Mr Gilbert said.
On Friday morning a duck race will begin at Darlington Point's new boat ramp to the old boat ramp, with funds going also being pledged to Country Hope.
The Redneck River Run will be launched from Figtree Park in Darlington Point from 7am on Friday.
