The Giants proved too good for Northern Knights, 10.17 (77) to 7.9 (51) at Sandringham.
There was a strong AFL Riverina representation for the Giants Academy, making up nine of the 23-man team.
Northern Jets teenager Jeromy Lucas starred with 28 disposals and two goals.
Liam Delahunty kicked three goals and had 11 touches, while Lucas Conlan kicked two goals and Shaun Driscoll was also named in the best.
It was a welcome victory for the Giants Academy, who had suffered losses to Brisbane Lions and Sydney Swans' academies to begin their campaign.
The Giants face Gippsland Power in their next game after Easter on Saturday, April 27 in Sydney.
