The Home and Away actress, who was diagnosed with Hodgkin's lymphoma at the age of 23, reportedly confirmed the news through her agent at the David Jones Spring/Summer launch on Wednesday night, the Daily Telegraph reports.
According to the Atelier Management agent, James is five months pregnant.
The actress attended Wednesday's night's event in a billowing, red Erdem frock, which hid any hint of a bump.
James and Melbourne Storm player Myles married in December 2011.
She received her cancer diagnosis in 2014, returning home to Australia from Los Angeles, where she had been auditioning for projects, to receive treatment.
The news came two years after her father, former Richmond player Stephen James, was diagnosed with non-Hodgkin lymphoma, which is incurable.
After 12 rounds of chemotherapy in six months, James, who has made guest appearances in Beaconsfield and Love Child, entered remission in 2015.
"It was amazing," she told Fairfax Media. "That's what you want. That feeling, it was huge relief and it was exciting that I was able to start a new chapter of my life and move on."
