Danny Roche and Andrew Vitucci will miss the clash meaning the side will be without one of their main weapons going forward as well as one of their rocks at the back.
"There is a big hole at the front and Andrew who can play multiple positions brilliantly, so it will be a bit of a loss," Yoogali SC coach Luke Santolin said.
"He (Roche) has been stood down for one match, but to be fair, his back was giving him a bit of grief, so we will find the silver lining and hope that he gets to rest up."
The ANU side is currently sitting at the top of the table level on points with O'Connor Knights, so it will prove a tough test for the Yoogali side, who are currently sitting just outside the top four.
The last time the two sides met, it was a 4-2 win for ANU, but the Yoogali side was coming into the game having played just two days earlier.
Santolin has full confidence that his side has what it takes to match it with the best in the competition.
"At our best, we fear no one," he said.
"I don't think, aside from the O'Connor game, I don't think there has been any side that has made us feel uncomfortable.
"Where we have been letting ourselves down is when we cop a goal or get an injury, or something happens, the pendulum has swung in the opposition's favour.
"We have spoken about wanting to change that and actually start bringing the momentum our way at those key moments, so we get another opportunity to do that on the weekend."

Coming into this weekend's game off the back of a 6-2 win over rivals Wagga City Wanderers, the Yoogali coach said his side are in a good headspace.
"Training last night was fantastic, the energy and the vibe was good, and the boys were really competing during the drills, and that is what a win can do," he said.
"We have come to terms with the fact that our numbers are our reality, and it isn't something that we can control.
"We will head in with the same 22 players that we know we can rely on, and we will go to war with people that we know we can trust."
Yoogali will also be using this weekend as a team-building exercise, according to Santolin.
"I think more than half of the squad is bringing up their wives, families or girlfriends, and we were staying overnight as a group," he said.
"We are trying to work on our culture off the field, which is normally one of our strengths."
The clash between Yoogali SC and ANU FC will kick off at 3pm on Sunday.
