NSW Health Infrastructure chief executive Rebecca Wark said feedback from staff and the community during consultation had "highlighted the opportunity to review and enhance the design".
"This has ensured it complements the unique Griffith Base Hospital site and the healthcare needs of local consumers; respecting the 1914 Walter Burley Griffin town-planning legacy, connecting key city sites along an axial link," Ms Wark said.
The design is meant to increase the amount of natural light into the building, and provide views over the area's landscape.
Feedback led to the building's scale being adjusted from the schematic to make it "more open and welcoming" and to improve the integration of the building on the hospital campus, while also retaining the same floor space for all departments.
"Consultation is continuing with staff and clinicians to further develop the design of each department located in the clinical services building," Ms Wark said.
The next step for the project will lodging a state significant development application with the NSW Department of Planning, Industry and Environment, with the application on public exhibition for 28 days.
The latest state of enabling work has begun at the hospital's site, beginning with the demolition of the former Matron's Cottage.
The hospital's re-development is estimated to be completed in 2025.
