Commencing on February 1, a 50-seater plane will offer travellers flights to and from the state capital every day of the week.
Qantas also announced special fares which will be available until December 20 or until sold out, which will see Griffith residents able to book one-way flights on the route from $125 when they begin next year.
The new route comes as part of an expansion to Qantas' regional network, with the airline announcing seven new routes across NSW, Victoria and South Australia today.
Council has welcomed the announcement, describing it as a "major boost for the region's economy".
Griffith mayor John Dal Broi said the decision to bring Qantas flights to Griffith is "recognition of Griffith as a growing regional city".

"Connectivity to Sydney is critical to our economy and lifestyle and Council is pleased residents and businesses will now have additional flight scheduling options," he said
QantasLink CEO John Gissing said the new route is an example of the company's efforts to encourage domestic travel.
"As the national carrier, we have an important role to play in driving tourism and supporting the industry's recovery," Mr Gissing said.
"Before COVID, more than 11 million Australians travelled overseas each year, so these flights will help convert some of these international trips into domestic holidays instead."
"We're also pleased to be able to offer Riverina residents more choice and competitive fares on these routes which have been monopolies for years."
The Qantas flights between Griffith and Sydney will be operated by their Q300 turboprop aircraft with upgraded interiors.
There are currently existing flights between Griffith and Melbourne operated by East-West Airlines, and flights between Griffith and Sydney operated by Regional Express Airlines.
Earlier this week, Regional Express announced flights between Sydney and Melbourne.
