Yoogali started the brighter of the two sides and had an early chance when Jaeden Andreazza was brought down in the box, but Luke Santolin was unable to convert the spot-kick with just five minutes gone.
The home side had a number of good chances following with Mason Donadel making a break down the wing before playing Andreazza in, but he was unable to put away the chance.
They didn't have to wait much longer to get some pay off for their early dominance with Jordan Serena popping up to tuck in a rebound after a looping shot from Donadel hit the crossbar and made it's way to the unmarked Serena.
Donadel provided once again soon after with a through ball finding Andreazza who was able to find the touch required to double his side's advantage.
Yoogali created another couple of chances late in the first half before O'Connor pulled one back with a penalty right before halftime.
They pulled level with another spot-kick in the second half, and after Jordan Baveresco was sent off for a second bookable offence, O'Connor found the winner late in the game after a long-range free-kick found it's way into the top corner.
Coach Sante Donadel was pleased with his side's performance an O'Connor team who he believes will be the team to beat.
"I thought we played well.
"It got a little bit scrappy when we went down to 10-men, but we created enough chances to win that game. The effort was defiantly their it was just our finishing letting us down today."
Donadel was pleased with his side's defence in open play with the only real chance the Knights created came from set-pieces.
"Luke (Armanini) didn't have to pull off a save all game," he said.
"I think their best player probably was their keeper. He pulled off a couple of great saves in the second half.
"It's just a case of you have to be able to score goals to win games."
Yoogali will look to rebound next weekend when they take on Canberra White Eagles at home.
