Starting the day just one stroke from the leaders, Herbert birdied three of his first four holes on Saturday to move to 10-under and take the outright lead.
He is one shot ahead of one of the overnight leaders and defending champion, Brett Rumford, who birdied the first hole to move to nine-under.
He was joined by Thorbjorn Olesen, who is two-under through the first three holes.
Former world No.1 Lee Westwood shared the lead with Rumford overnight but has failed to improve on his 36-hole score of eight-under.
New South Welshman James Nitties is the big mover early in the third round.
He started the day two-under and has picked up five shots through 11 holes to be sitting just three shots from the lead at seven-under.
Only the top 24 golfers will qualify for the final-round match play component of the innovative tournament, where players will go head-to-head in a series of six-hole shootouts.
The top eight golfers entering the final day earn a precious bye in the first round of match play.
If players are tied after their six-hole match play battle, their clash will be decided on a 90m 'shootout' hole.
